Midbrain dopamine (DA) neurons discharge in a single-spike and burst discharge mode in vivo. The discharge rate and the prevalence of the burst-discharge pattern increases in an age-dependent fashion. DA neurons are also spontaneously active in vitro. However, the in vitro discharge rate and pattern is very different from the pattern that is observed in vivo. In vitro, the discharge pattern is almost exclusively limited to a regular single-spike or pacemaker mode. Only rarely have DA neurons been observed to display burst-discharges in the midbrain slice preparation. The action potential discharge rate increases, and the discharge pattern becomes more regular in an age-dependent fashion during postnatal development. Increases in intracellular cAMP concentration elicited responses characterized as excited, inhibited, or biphasic (excited and inhibited). An increase in intracellular cAMP elicited by forskolin or 8-Br-cAMP was shown to change the discharge pattern of immature DA neurons discharging in an irregular discharge pattern to a pacemaker-like discharge pattern similar to that observed in more mature DA neurons. Increases in intracellular cAMP caused depolarization and in some cases the DA neurons switched to a burst-discharge mode. The effects mediated by 8-Br-cAMP and forskolin on DA neurons were found to have an age-dependent component. The age-dependent aspect of the responses suggests that the adenylate cyclase system may still be undergoing developmental changes during the postnatal period.

This is the first report of an endogenous second messenger causing a DA neuron to switch from the single-spike mode to the burst-discharge pattern. In SNC DA neurons, the c-AMP-mediated effects were observed as follows: (1) membrane depolarization, (2) increased conductance, and (3) decreased AHP amplitude. Evidence strongly suggests that excitation and most inhibitory responses are both mediated by depolarization. There is evidence to indicate that inhibition was, in some instances, mediated by hyperpolarization. The exact channels that are modulated by cAMP remain to be elucidated, however, preliminary evidence suggest the SK-type calcium-mediated potassium channel (IK(Ca)) is involved.
