The suprachiasmatic nucleus (SCN) of the hypothalamus is a master circadian pacemaker. It is divided into a core and a shell on the basis of differences in chemoarchitecture, cytoarchitecture, and the pattern of afferent innervation. However, there is no unifying definition of the core and shell and, consequently, the specific functions of the individual subdivisions are unknown. The SCN has dense intrinsic circuits and efferent projections to various diencephalic targets, but their precise organization, or whether they differ for the core and shell, is not known. An understanding of the anatomical features of the SCN is important because its efferent projections relay rhythmicity to effector sites while its local circuits are thought to synchronize neuronal oscillators. The present study investigates the local and efferent connections of the SCN using three tracers, Cholera Toxin beta subunit (CTB), FluoroGold (FG), and pseudorabies virus (PRV) Bartha, a neurotropic herpesvirus which is used as a retrograde transneuronal tracer. We hypothesized that the core and shell differ in the organization of their local and efferent connections, such that the pattern of retrograde labeling corresponds precisely to that of peptide phenotype and afferent innervation. First, major SCN targets were injected with tracer and the retrograde labeling compared to vasopressin and neuropeptide Y immunoreactivity on adjacent sections. These peptides delineate subdivision boundaries, vasopressin being expressed in shell neurons and neuropeptide Y in afferent terminals in the core. Second, two targets, one innervated preferentially by the shell and one by the core, were injected with virus and the survival period varied to follow the transynaptic passage of virus through SCN circuitry. The results indicate that the core and shell have different terminal projections both within and outside the nucleus. The pattern of retrograde labeling improves our definition of the core and shell and illustrates how consistent it is across different anatomical features. This implies that the subdivision of the pacemaker is indeed a functional one, and that it probably involves the synchronizing and output components of the circadian timing system.
