The purpose of this investigation was to evaluate the age-altering effects of uncoupling protein 2 (UCP2) on oxidative stress in tissues by using female C57BL/6J mice models. Mice that overexpressed or lacked the target gene of UCP2 were compared to wild type ad libitum and wild type caloric restricted to determine the differences in the level of oxidation stress at 8, 19 and 26 months of age. Results showed that the absence of the UCP2 gene increased lipid peroxidation levels in middle-aged mice, but there were no consistent evidence of UCP2 overexpressors decreasing the production of reactive oxygen species (ROS). In brief, there were no sufficient evidence to state that ROS regulations by UCP2 induce life span prolongation effects; however, overexpression of UCP2 does play a limited role in regulating lipid peroxidation in middle age.
