Steroid hormone biosynthesis is dependent upon the synthesis of steroidogenic acute regulatory protein (StAR). Regulation of the adrenal hormone, aldosterone, is critical for regulating blood pressure, and the main regulator of aldosterone is angiotensin II (Ang II). Therefore, Ang II regulation of StAR is key to aldosterone production. I have identified the DNA elements and transcription factors important for Ang II activation of the human StAR gene. I provide the first molecular characterization for hStAR activation by showing that SF-1, YY1, and CREMalpha/gamma are recruited to the proximal StAR promoter after Ang II treatment. Further, I show that COUP-TF I increases the basal activity of the StAR promoter and binds simultaneously with GST-SF-1 to the functional SF-1 site in the StAR proximal promoter. I propose that COUP-TF I regulates basal StAR transcription and that YY1, SF-1 and CREM alpha/gamma regulate the Ang II response.
