Mobile source air toxics present a significant public health concern: (1) they are carcinogenic and toxic; (2) exposures are widespread due to their integration within communities; and (3) their lipophilicity may place lactating mothers and their infants at particular risk. The purpose of this dissertation is to further define this public health threat in two related ways. The first is to characterize mobile source pollutant concentrations and source contributions within an urban parking garage as a potential "hot spot". The second is to develop and test methods for measuring mobile source air toxins in human milk.

520 $a The first objective was achieved using time-resolved measurement methods to characterize air pollutant levels within an urban parking garage and assess the differential impact of sports utility vehicles relative to sedans. Carbon monoxide (CO) and particle-bound polycyclic aromatic hydrocarbons (pPAH) were measured with direct reading instruments. Volatile organic compounds (VOCs) including 1,3-butadiene, methyl tert-butyl ether (MTBE) and benzene were measured in 30 min intervals using sorbent tubes. Vehicle volume and type were evaluated using a video recording. Sampling was conducted 8-h/day (7:00 am to 3:00 pm) for 24 days from June 24 to July 17, 2002.
520 $a The second objective was achieved by methods optimization and measurement of VOCs and PAHs in human milk. Laboratory studies using unhomogenized bovine milk were conducted to assess the potential for VOC losses during sampling and optimal temperature and duration for extraction. Using the optimized methods from above, human milk was collected and analyzed from two sources: a North Carolina milk bank (n=5) and three women living in homes with no smokers in inner-city Baltimore, MD (n=8). Milk VOCs (i.e., MTBE, chloroform, benzene and toluene) were extracted using head space solid phase micro extraction (HS-SPME) whereas PAHs (i.e., fluorene, phenanthrene, fluoranthene, pyrene and benzo[a]pyrene) were analyzed by solid and liquid phase extraction. Both VOCs and PAHs were analyzed by isotope dilution GC/MS/SIM. For the Baltimore samples, 24-h VOC air concentrations were measured by active sampling and thermal desorption GC/MS/SIM over each of the three days of milk collection. Results from the urban garage "hot spot" study indicated garage traffic median volumes of 23 and 48 counts/hr for SUVs and sedans on weekdays and 2 and 4 counts/hr on weekends, respectively.
