Ontogeny of the lymphoid organs and the immune response to Edwardsiella ictaluri in channel catfish, Ictalurus punctatus, fry and fingerlings. - 170 p.

Channel catfish leukocytes demonstrated cytoplasmic staining patterns very similar to mammalian leukocytes when stained for acid phosphatase, a-napthyl butyrate esterase, b-glucuronidase and Sudan Black B. Monocytes, macrophages, neutrophils and possibly immature lymphocytes were present in the channel catfish renal hematopoietic tissue at hatch. Surface immunoglobulin positive (sIg+) cells were first present on days 7 and 10 post-hatch in the anterior and posterior renal hematopoietic tissues, respectively. SIg+ cells were first present in the liver and heart on day 10. Monocytes, macrophages, and possibly immature lymphocytes were present in the thymus at hatch. Intermediate stage thymocytes were present on day 7, and sIg+ cells appeared on day 10 in some fry, were absent and reappeared again on day 21. Splenic monocytes, macrophages and neutrophils were present on day 1, but sIg+ cells were not present until day 14. Thymic differentiation was not apparent until day 21, and the spleen demonstrated minimal lymphoid organization on day 21. Morphologic data supports the functional data. In two separate studies, specific antibody titers were detected in fry that were exposed to live bacteria at 3 and 4 weeks post-hatch. In the study involving secondary exposures, a heightened secondary response was first demonstrated by fry that received a primary exposure at 4 weeks post-hatch. This study indicates that channel catfish are not capable of generating a specific immune response before three weeks of age.
