Chemical synaptic transmission is an essential process that enables neurons to communicate with each other through release of neurotransmitters. The mechanisms of neurotransmitter release and clearance are tightly regulated and precisely controlled through different sets of modulators. Although much about how this regulation is mediated is well known, there are still many unresolved questions. Moreover, as organisms age, the synaptic integrity declines due to molecular changes that may contribute to the onset of neuropathological disorders. Cholinergic synaptic transmission is essential for proper memory, cognitive and locomotion performance. Similarly, the dopaminergic system regulates movement and various brain functions, including motivation and reward. Both systems are interconnected, and through aging, their degeneration exacerbates the occurrence of neurodegenerative diseases. The objective of the study was to identify how changes in VAChT expression affects the acetylcholine levels and the ACh-linked behaviors during aging of Drosophila melanogaster. We also aimed to characterize a robust a-synuclein Drosophila model for PD. Given that cholinergic and dopaminergic dysfunction is a characteristic feature of Alzheimer's Disease and Parkinson's Disease, it is expected that the proposed studies will provide new insight into the changes in neurotransmission observed in the mentioned disease states and may contribute to the development of novel therapeutic strategies.
