This dissertation investigates the role of Mucosal-Associated Invariant T (MAIT) cells in colon cancer and their transcriptional regulation governing activation and effector functions. In Chapter 2, using a RAG1 knockout background, it is demonstrated that peritumoral injection of MAIT cells inhibits tumor growth in an MC38 mouse model. This intervention is associated with a notable increase in proinflammatory cytokines and cytokines modulating innate cell responses within the tumor microenvironment.Chapter 3 delves into the role of transcription factors STAT1, STAT3, STAT5, and HIF1α in regulating MAIT cell activation, effector functions, and glycolytic metabolism. These findings lay a foundation for future research aimed at integrating the study of transcriptional regulation of MAIT cell functions in disease models, particularly in colon cancer. The insights gained from this dissertation can be extended to explore broader aspects of MAIT cell functionality in various immune-related diseases.
