A mathematical model of CHO (Chinese Hamster Ovary) cells has been developed to predict the general growth characteristics of cells under a wide range of external conditions existing in tissue culture bioreactors. The model predicts the growth rate of the CHO cells and the cellular macromolecular composition in response to perturbations in the external pH, levels of glucose, glutamine, lactate, ammonia and other amino acids. The model has been tested with available literature data and original chemostat experiments and T-flask experiments. The model specifically included the detailed structure necessary to account for the activity and regulation of the two major carbon sources for cultured mammalian cells--glucose and glutamine. For the growth of CHO cells in suspension, the apparent saturation constant for glucose was experimentally determined to be about 0.14 mM, and the apparent saturation constant of growth on glutamine was about 0.10 mM. The model is capable of predicting both values. In particular, the model is capable of predicting the intracellular pH in CHO cells. The intracellular pH is a major determinant of many intracellular biochemical processes, and the major metabolic by-products such as lactate and ammonia, inhibit cell growth by disturbing the optimal intracellular pH for cell growth. The model predicts the metabolism of glucose and glutamine and consequently the production of lactate and ammonia. It included the detailed structure of intracellular pH regulation by the Na/H antiport, disturbance by lactate and ammonia, proton leakage, and intracellular buffering capacity. This model is the first to integrate intracellular pH prediction with a complete model of cell growth, allowing investigators to predict a priori a wide range of growth response to environmental perturbations.
