Early detection and therapeutic intervention of deep tissue hypoxia can prevent severe ischemic clinical pathologies, such as tissue necrosis, gangrene, organ dysfunction, or hypertension. Photoacoustic (PA) imaging is a hybrid imaging modality that uses optical excitations to generate acoustic waves allowing imaging at optical resolutions with imaging depths on par with clinical ultrasound modalities. The use of optical excitations allows probing of structural, functional, molecular, and kinetic information through thermoelastic expansions facilitated by optical chromophores within the imaged media. Examples of these include endogenous agents like hemoglobin, lipid, melanin and water or a variety of exogenous contrast agents. By acquiring images at multiple wavelengths, spatial maps of chromophore distribution can be retrieved. However, biological tissues are highly complex with attenuation of light being the primary limiter for accurate quantitative imaging of deep tissue (> 2 cm). Therefore, the objective of this work is to optimize spectral PA imaging for deep (∼ 5 cm) highly complex biological tissues. This is explored via two main aspects - optimizing the choice of wavelengths used to probe endogenous chromophores and characterize a novel biocompatible exogenous contrast agent. To demonstrated clinical translatability, these results are used to develop a portable integrated PA probe capable of monitoring deep tissue arterial blood perfusion.
