The foundational question of my dissertation research was "how do ~12-hour biological rhythms intersect with proteostasis?". I begin with a review of ultradian chronobiology (Chapter 1) that will equip the reader with the relevant background information to understand the following two chapters. Next, I present our study linking the nuclear speckle's ~12-hour rhythms of liquidliquid phase separation dynamics to ultradian rhythms of proteostasis gene expression (Chapter 2). Then, I report our research establishing the liquid-liquid phase separation dynamics of the nuclear speckle as a druggable target to modulate the cell's ability to maintain a balanced proteome (Chapter 3). Finally, I provide concluding remarks that I hope leave the reader excited for the future of nuclear speckle research. This dissertation coherently explains the importance of ultradian chronobiology to organismal health, and details how the simple aforementioned question led to the establishment of the nuclear speckle as a druggable target to boost cellular proteostasis.
