Emiliania huxleyi, the dominant species of coccolithophores in modern oceans, contributes prolific quantities of carbon fixation through calcite biomineralization. This species plays a prominent role in the ocean's biogeochemical cycles and its unique production and shedding of carbonate coccoliths may serve as inspiration for carbon capture and sequestration technologies. This thesis includes three original research studies that set out to discover genes, elements and molecules involved in E. huxleyi biomineralization. The first study employed cell penetrating peptides to overcome the coccosphere barrier and deliver gene specific peptide nucleic acids to viable E. huxleyi cells. Direct delivery of peptide nucleic acids represents a useful synthetic biology tool to further explore coccolithophore biology. In the second study, coccolith elemental incorporation was mapped using high resolution x-ray fluorescence, revealing trace elements embedded in and around the coccolith. Based on elemental maps of sulfur, we hypothesize that sulfur plays and expanded role in the global sulfur cycle and may play a direct role in E. huxleyi biomineralization. In the final study, chemical spectra of single live cells were measured by synchrotron FTIR, demonstrating the capability of differentiating between calcified and decalcified cell types. This works demonstrates a useful result for exploration of E. huxleyi calcification heterogeneity in response to chemical, genetic and environmental perturbations.
