Diabetes is a chronic metabolic disorder that presents a major worldwide health problem. It is characterized by an absolute or relative deficiency of insulin secretion and/or insulin action associated with chronic hyperglycemia and disturbances of carbohydrate, lipid, and protein metabolism. The persistent hyperglycemia plays a key role in the development of long-term complications that including retinopathy, nephropathy, coronary heart disease, peripheral vascular disease and cerebrovascular disease, such as peripheral neuropathy and central nervous system's diseases, e.g. dementia and Alzheimer's disease.There are several mechanisms by which hyperglycemia leads to Alzheimer's disease. They include impaired glucose metabolism and insulin resistance in the neural cells; accumulation of toxic molecules, such as amyloid beta that leads to phosphorylation of tau-protein- all resulting in mitochondrial dysfunction and oxidative stress, leading, eventually, to apoptosis and cell death.Various medications that are used to treat diabetes do not affect equally all patients, and some of them cause various side effects. As a consequence, there is an increased interest during recent years in new anti-diabetic materials, isolated from natural sources.The GTF, Glucose Tolerance Factor, is a dietary agent extracted from brewer's yeast, which increased glucose transport into various cells and reversed glucose intolerance in diabetic animals and humans. Previous studies done in our laboratory on rats showed that oral treatment with GTF decreased blood glucose and lipids and potentiated insulin action in both types of diabetes.The aim of our study was to analyze the effects of high glucose conditions and GTF treatment on pheochromocytoma (PC-12) cells, as an in vitro model for neural cells. We examined the effects of GTF on cell viability, cell death and caspase-3 activity in those cells.PC-12 cells were grown for 48 or 60 hours in normal medium (24.75 mM glucose) and in high glucose medium (5 fold or 7.5 fold normal glucose levels) with or without addition of GTF.GTF was added for 24 hours. MTT (3-(4,5-dimethyl-2-yl)-2,5-diphenyl-2H tetrazolium) assay was used to assess cell proliferation; Determination of cells death was done by a fluorescence FACS (fluorescence-activated cell sorting) assay; and caspase-3 activity was measured by spectrophotometer set to 405 nm light absorption. We observed a significant decrease in PC-12 proliferation in high glucose conditions of 88.8% and 64.6% of normal rate (for x5 and x7.5 glucose compared to normal rate, respectively).Addition of GTF (1mg/ml) to the cells grown in a 7.5 fold glucose medium, increased the proliferation rate to 79.2%.We also observed significant increase in apoptotic cells death in glucosex7.5 conditions of 2.24 fold compared to normal rate.Addition of GTF (0.25mg/ml, 0.5mg/ml, 1mg/ml and 2mg/ml) to the cells grown in a 7.5 fold glucose medium, decreased cell apoptotic death to about 1.5 fold of normal rate (by 67% less death than in high glucose conditions).We also observed significant increase in caspase-3 activity in glucosex7.5 conditions of 128% compared to normal rate.Addition of GTF at 2mg/ml or 10mg/ml to the cells grown in 7.5 fold glucose media compared to normal, decreased caspase-3 activity rate to 86% and 70% of compared to normal rate, respectively.In conclusion, our study demonstrates that high glucose conditions decreased cells viability, induced apoptotic process and elevated caspase activity. Addition of GTF to high glucose medium decreased the occurrence of apoptotic events and improved cells proliferation.

520 $a סכרת היא מחלה מטבולית כרונית אשר מוכרת כיום כבעיית בריאות עולמית. מחלה זו מאופיינת על-ידי חוסר מוחלט או חלקי בהפרשת אינסולין או בפעילותו אשר מתבטא בעליה כרונית של גלוקוז בדם )היפרגליקמיה( והפרעה בחילוף חומרים של פחמימות, שומנים וחלבונים. המצב הכרוני של היפרגליקמיה גורם בטווח הארוך לסיבוכים רבים: רטינופתיה, נפרופתיה, מחלות לב, מחלות במערכת הוסקולרית הפריפרית, כולל נוירופתיה. נוירופתיה יכולה להיות פריפרית, ולהתבטא באבדן תחושה, או מרכזית, ולהתבטא במחלות נוירודגנרטיביות, כמו פרקינסון, דמנציה ומחלת אלצהיימר.המנגנונים שבהם היפרגליצמיה מובילה למחלת אלצהיימר הם רבים ומגוונים, וכוללים פגיעה במטבוליזם הגלוקוז, ועמידות לאינסולין בתאי הנוירון; הצטברות מולקולות רעילות, כמו עמילואיד-ביטא, המובילה לזרחון של חלבון טאו- כל המנגנונים הנ"ל מובילים לפגיעה בתפקוד המיטוכונדריה ועקה חמצונית, המוביליםד לאפופטוזיס ומוות תאי.תרופות שונות המשמשות לטיפול במחלת הסכרת אינן משפיעות במידה שווה על כל החולים, ולחלקן אף תופעות לוואי שונות. כתצואה מכך, בשנים האחרונות גברה התעניינות במחקר של חומרים אנטי סכרתיים חדשים, בעלי פעילות ביולוגית המבודדים ממקורות טבעיים.F,Factor Tolerance Glucose,הינו חומר טבעי המופק משמרים, בעל פעילות אנטי סכרתית. החומר מגביר את כניסת הגלוקוז למגוון תאים ומשפר סבילות לגלוקוז בחיות ובני אדם סכרתיים. בניסויים שנערכו בחולדות סכרתיות במעבדתנו, נמצא כי GTF גורם לירידה מיידית של רמת הגלוקוז והשומנים בדם, ומגביר את פעילות האינסולין בשני סוגי הסכרת.מטרת מחקרנו הנוכחי הייתה לבחון את ההשפעה vitro in של ריכוזי גלוקוז גבוהים על שגשוג, על מוות תאי ופעילות קספאז-3 .בתאי נוירון מסוג פאוכרומוציטומה (12-PC,)והשפעת GTF המוסף למדיום גידול המכיל ריכוזי גלוקוז גבוהים על מניעת תופעות אלו.התאים גודלו במשך 84 או 06 שעות במדיום גידול נורמלי המכיל mM 75.24 גלוקוז, ובמדיום גידול המכיל ריכוז גלוקוז גבוה של mM 75.123 או mM 6.185,עם או ללא תוספת GTF. GTF הוסף למשך 48 שעות.כדי לבדוק את התרבות התאים, נעשה מבחן MTT ;כדי לקבוע מוות תאי, נעשה שימוש בשיטת בליעה פלורוסנטית FACS,ופעילות קספאז-3 נמדדה באמצעות בליעת אור של תוצר הפירוק )pNA מפורק( בספקטרופוטומטר באורך גל של 405nmבמחקרנו נמצא כי הוספת גלוקוז גבוה לתאים גרמה לירידה מובהקת בשגשוגם ב-%4.44 ו-%0.08 בהשוואה לגידולם בתנאים נורמליים )בגלוקוז פי 5 ופי 5.5 בהתאמה(. הוספת GTF העלתה את שגשוג התאים שגודלו בגלוקוז פי 5.5 ל-%4..5 לעומת גידולם בתנאים נורמליים. עוד נמצא במחקר, כי הוספת גלוקוז פי 5.5 מהנורמה העלתה באופן משמעותי מוות תאי אפופטוטי, פי 48.4 מהמוות התאי הנצפה בתנאי גידול של גלוקוז ברמה נורמלית. תוספת GTF( בריכוזים 45.6,5.6, 1 ו-4 מ"ג/מ"ל( לתאים שגודלו בתנאי גלוקוז גבוהים )פי 5.5 )גרמה לירידה במוות התאי עד כדי פי 5.1 מהמוות התאי הנצפה בתנאי גידול נורמליים )עד %05 פחות מוות תאי מתנאי גידול של גלוקוז גבוה פי 5.5 ללא תוספת GTF .)בנוסף, מניסויים ראשוניים של בדיקת פעילות קספאז-3, נצפתה עליה משמעותית בפעילותו בתנאי גלוקוז גבוהים פי 5.5 מהנורמל, עד כדי %144 פעילות האנזים לעומת פעילותו בתנאים נורמליים. תוספות של GTF( בריכוזים 4 ו-16 מ"ג/מ"ל( לתאים שגודלו בתנאי גלוקוז גבוה פי 5.5 מהנורמה, הורידו את פעילות קספאז-3 עד כדי %40 ו-%56 מפעילותו בתנאי גידול נורמליים, בהתאמה.לסיכום, ממחקר זה ניתן להסיק כי גידול נוירונים בתנאי גלוקוז גבוה - מזיק לתאים, מוריד את השגשוג, מעלה תהליכי אפופטוזיס ומגביר פעילות קספאז. תוספת GTF מורידה את פעילות הקספאז ומעלה את השגשוג בתאי נוירונים.
