Photoactivatable therapeutics are targeted light-activated drug delivery systems composed of a therapeutic cargo appended to a photoresponsive moiety, which releases the cargo upon illumination. These targeted therapeutics have the potential to diminish toxicity and improve efficacy of the therapeutic cargo due to the temporal and spatial control provided by external light controlled therapeutic release. The introduction of this dissertation describes the precedent set by clinically approved photodynamic therapy and currently available photoresponsive moieties. Together, this background contributes to the proposed pre-clinical development plan for photoactivatable therapeutics.Vitamin B12, or cobalamin (Cbl), has shown exciting potential as a photoresponsive moiety with inherent photoresponsivity and minimal toxicity. Herein, two Cbl-based photoactivatable therapeutics encapsulated in red blood cells (RBCs) are developed. In vivo studies of a Cbl RBC chemotherapeutic and anti-inflammatory drug demonstrate localized delivery and efficacy comparable to standard of care respectively. Together, the advances contribute to the pre-clinical development plan for Cy5-Cbl photoactivatable therapeutics proposed in the conclusion. Overall, photoactivatable therapeutics, and Cy5-Cbl therapies specifically, are not far from first in human trials.
