How crop plants respond to stress in their physical environment is crucial to productivity. The response of plants to drought stress is mediated by changes in gene expression, but the molecular basis for plant tolerance to this condition is not completely understood. The methodology of differential display has been used in several biological systems that involve changes or differences in gene expression. We improve the differential display technique to detect differences in mRNA transcripts between irrigated melon plants and eight-day non-irrigated plants. The percentage of false positives was reduced by optimizing the RT-PCR conditions, by increasing the running time of the sequencing gel, and by running six replications. We obtained fewer transcripts for the plants under stress as a direct influence of the stress itself. Most of the differential display transcripts were downregulated as a possible response mechanism of the plant to save energy. A total of 18 bands were differentially expressed, 14 of which were present only in irrigated plants while four were found to be present only in non irrigated plants. We identified five cDNA clones that gave a positive signal on the reverse northern blot, all of them from irrigated plants. We suggest a possible role of these transcripts in the response of plants to drought conditions.
