A relationship between the arabinose content of hemicellulose in pulp, and moisture adsorption, has recently been observed. It was therefore hypothesized that the selective removal of arabinose from pulp by treatment with alpha-L-arabinofuranosidase would enhance the moisture retention properties of pulp. The objectives of this research were to develop microbial sources for alpha-L-arabinofuranosidase and xylanase, to treat unbleached kraft pulp with crude preparations of these enzymes and to assess the effect the enzyme treatments have on the hygroscopic properties of the pulp. Heteroxylans are a major component of corn fiber, making it a potential source of fermentable carbohydrates for fuel production. Therefore, an additional objective of the research was to investigate the enzymatic saccharification of corn fiber.

520 $a alpha-L-arabinofuranosidase was purified from Aureobasidium pullulans using a sephacryl-HR-200 gel permeation column and a DEAE-Sepharose anion exchange column, resulting in 39% total recovery in enzyme activity. Xylanase was partially purified from Trichoderma longibrachiatum with 43% recovery in total enzyme activity and a purification factor of 19.8. Corn fiber was treated with crude preparations of both enzymes, and the amounts of carbohydrate produced were comparable to the amounts produced by conventional acid hydrolysis. Unbleached kraft pulp was treated with concentrated crude preparations of alpha-L-arabinofuranosidase and xylanase, obtained by ultrafiltration of the respective culture supernatants. The effect of these treatments on moisture adsorption was investigated by incubation of the pulp samples in a constant humidity chamber. Treatment with alpha-L-arabinofuranosidase provided enhanced moisture gain resistance with no yield loss, both of which are of major importance in paper manufacturing.
