Sulfadimethoxine (SDM), an antibacterial drug in human and veterinary medicine, has been proposed for use in fish husbandry. Pharmacokinetic and metabolic data for SDM in aquatic biota are sparse; therefore, assimilation, kinetics, tissue distribution, metabolism and pathways of elimination of SDM have been measured in channel catfish (Ictalurus punctatus). These studies support drug development for the widely cultured channel catfish and contribute to the understanding of drug pharmacokinetics in fishes.Subjects--Topical Terms:

520 $a Mature (400-600 g/35-40 cm) and immature fish (30-40 g/15-20 cm) were dosed with 40 mg/kg SDM. The vascular clearance and tissue distribution of SDM were measured after single and multiple intravenous and oral doses of $\ sp{14} $\ sp{35}
520 $a SDM tissue burdens in channel catfish increased with the number of doses administered. The elimination rate constant of SDM from individual tissues did not increase, however, as tissue burdens increased.
520 $a The biphasic blood elimination of SDM after intravenous dosing conformed to a two compartment model. The initial ( $\ alpha $) elimination phase was very rapid (0.09 hr), and may be associated with the low plasma protein binding of SDM in channel catfish. The final ( $\ beta $) elimination half-time (13-hr) classified SDM as a long acting sulfonamide in channel catfish. The calculated pharmacokinetic parameters which described SDM distribution and elimination after multiple-dosing agreed with values determined for a single dose in mature fish. Younger fish, however, had a decreased time for total body clearance of the drug.
520 $a Biliary elimination in fish is the major pathway of excretion of non-polar compounds of SDM, whereas in mammals urinary elimination is preponderant. Although the rapid clearance of SDM from the plasma of channel catfish after dosing leads to low plasma concentrations of SDM, the longer $\ beta $ elimination rate, which is similar between catfish and mammals, allows the establishment of specific dosing regimens to obtain therapeutic tissue levels in the aquaculture industry.
