Plants are immobile organisms, unable to escape unfavorable conditions. Instead, plants have developmental plasticity to respond to their environment. Light, which provides the energy for growth and reproduction, is the principal feature affecting a plant's survival. Plants sense their light environment through various wavelength-specific photoreceptors, which initiate intracellular signaling processes that adaptively modulate gene expression and development. Phytochrome A (phyA) is the best-understood photoreceptor although the biochemical events linking light perception to plant response are not understood.

520 $a Far-red control of hypocotyl elongation by phyA served as the basis for a genetic screen to identify novel phyA signaling components. Seven mutants were identified. The recessive mutant fhy4-1 revealed a novel locus. Its far-red light specific long hypocotyl phenotype indicates involvement in phyA signaling. Interestingly, fhy4-1 is also seedling lethal under white light, with a lack of chlorophyll and carotenoid accumulation. Scanning electron microscopy (EM) revealed a slightly lowered stomatal density for far-red grown fhy4-1 and that a cell elongation defect produces the elongated hypocotyl. Transmission EM showed that the plastids of white and far-red grown fhy4-1 cotyledons are not fully differentiated. Microarray analysis did not identify any significantly affected major pathways, though a smaller proportion of the genome shows significant far-red light regulation for fhy4-1 than for phyA mutant seedlings, reflecting its comparatively mild phenotype and possibly placing it downstream in the phyA pathway. Mapping and TAC-based complementation tests defined FHY4 in a narrow region of Chromosome 2, but candidate gene sequencing and insertion line screening did not reveal the nature of the fhy4-1 mutation.
520 $a A null-like dominant negative mutant, phyA-300D, was also characterized which has far-red specific elongated hypocotyls and closed cotyledons. The mutation, a valine to methionine change in PHYA at codon 631, does not compromise the dimerization capacity or spectral activity of the mutant phyA monomers. The corresponding PHYB mutation did not produce a dominant negative phenotype, indicating specificity of this mutation for the phyA pathway.
