The suprachiasmatic nuclei (SCN) of the hypothalamus are the principal site of circadian rhythm generation. Their 24-hour rhythm is under the regulation of the daily light-dark cycle as well as under autonomous pacemaker activity. In the present study we used in vivo and in vitro approaches to examine the role of polysialic acid (PSA) and its protein carrier, neural cell adhesion molecule (NCAM), in the regulation of light- and behavior-induced phase resetting of the clock. PSA-NCAM is well-identified as the principal regulator of axon sprouting and path finding, synaptogenesis, and cell-cell communication, among others, in the developing nervous system. In the adult brain, only select few plastic regions such as the hippocampus, olfactory bulb, and the hypothalamic SCN express PSA-NCAM and thus retain the capacity to undergo structural reorganizations. Our immunoblot analyses provide the first evidence of its kind for a daily and an endogenous rhythm of PSA and NCAM expression in the adult SCN of Syrian hamsters. Additionally, microscopic observations revealed that light-induced PSA is associated with specific markers of SCN neuronal activity, namely the Fos protein, calbindin D-28K, and the BDNF receptor, TrkB, indicating that PSA-NCAM may modulate photic signaling processes within the SCN cells. Finally, the removal of PSA from the SCN or the thalamic intergeniculate leaflet (IGL) by a specific endoneuraminidase (endo N) resulted in the suppression of light-induced and the potentiation of behaviorally- and 8-OH-DPAT-stimulated phase shifts. These data are intriguing in that the effect on the two inputs is opposite in direction. This difference could represent the mutually antagonistic relationship between photic and non-photic stimuli and also could reflect the permissive role of PSA on cell-cell interactions. In general, these results provide sound evidence implicating PSA-NCAM as an integral modulator of photic and non-photic signaling necessary for normal phase-resetting of the adult circadian clock.
