Genetic approaches have been highly successful in identifying pacemaker components in Drosophila especially in the past decade. Several components of the central pacemaker have been identified but the function of many of these remains obscure. The timeless gene product is essential for behavior and molecular oscillations. It has been suggested to play an important role in transmitting photic information to the pacemaker. Spectral and genetic evidence supporting a role for TIMELESS (TIM) in circadian photoresponses is presented. TIM heterodimerizes with PERIOD (PER) and the PER-TIM dimer is believed to be the central pacemaker species. Evidence for involvement of TIM and/or the PER-TIM dimer in amplifying PER oscillations both post-transcriptionally and post-translationally is presented.

520 $a Temporal phosphorylation of PER is a major feature of the Drosophila molecular pacemaker. Genetic and biochemical data implicate the DOUBLETIME (DBT) kinase as a central pacemaker component. The observations described here suggest a role for DBT in regulating the stability of PER as well as its involvement in photic coupling of the pacemaker. Collectively, the data suggests a dynamic interplay between TIM and DBT in regulating pacemaker kinetics. Thus, TIM stabilizes PER by sequestering it in a tight complex and DBT destabilizes PER by phosphorylating it. Circadian time is likely determined by the relative abundance of free PER and that bound to TIM.
