Low back pain continues to be a significant clinical and economic problem that causes major disability in our society. Although the exact mechanism of low back pain is unknown, an important factor is believed to be intervertebral disc degeneration.

520 $a Low back pain continues to be a significant clinical and economic problem that causes major disability in our society. Although the exact mechanism of low back pain is unknown, an important factor is believed to be intervertebral disc degeneration.
520 $a Current clinical treatments include non-surgical (conservative) and surgical approaches. Conservative treatments will relieve symptoms for the majority of back pain patients, however a large number of patients still remain who require surgical intervention due to the failure of conservative treatment. It is widely known and accepted that results from current surgical techniques typically benefit patients for less than 5 years and may result in unfavorable long term sequelae. Additionally, a separate joint disease may be created adjacent to the original diseased joint due to the load sharing of adjacent interdependent motion segment components.
520 $a Much effort, funds, and personnel have been invested in modern tissue engineering technique in order to find a new treatment modality for the disease, but researchers in the field are working with the unfavorable biological and metabolic environment of the intervertebral discs. Therefore, a useful modality from this research is still decades away.
520 $a Age-related crosslinks were proposed as a product of the aging process, by several authors of literature, since these age-related crosslinks were found to be increased in degenerated or aged specimens, such as degenerative intervertebral discs. The hypothesis was proposed, without experimental verification, that these crosslinks are responsible for the brittleness and reduced fatigue resistance in the degeneration process. However, our point of view, based on our pilot study, is that the increase of age-related crosslinks provides additional structural enforcement to protect against tissue degradation.
520 $a Our hypothesis has led to the development and testing of a method to augment the extracellular matrix of collagen in intervertebral discs through the usage of exogenous crosslinker. The specific aims of these efficacy investigations were to determine the effects of the crosslink augmentation on spinal motion segment stability and poroelastic properties of the intervertebral discs.
