Using zebrafish as a model, the present study aims at investigating the role of activin in the gonadotropin regulation of final oocyte maturation, and its interactive relations with other local factors including epidermal growth factor (EGF), transforming growth factor-α (TGF-α) and gonadotropin releasing hormone (GnRH). An <italic>in vitro</italic> follicle incubation and a novel primary follicle cell culture system were used in the present study. The regulation of gene expression of activin and its receptor was analyzed by a semi-quantitative RT-PCR assay.

520 $a The results showed that both activin βA and activin type II receptor are expressed in the zebrafish ovary and recombinant goldfish activin B significantly stimulated zebrafish oocyte maturation <italic>in vitro</italic>. The effect of activin could be abolished by follistatin, an activin binding protein. Interestingly, follistatin also blocked the stimulatory effect of gonadotropin (hCG) on the oocyte maturation, suggesting that gonadotropin may exert its action in the ovary by activating the ovarian activin system. Taken together, the lines of evidence strongly suggest a central role for activin system in the endocrine and paracrine regulation of zebrafish ovarian functions. The action of activin in the ovary involves <italic>de novo</italic> protein synthesis at both the transcriptional and translational levels because its effect could be blocked by both actinomycin D and cycloheximyde. GnRH is a well-known neuropeptide that plays a vital role in controlling pituitary gonadotropin secretion. However, substantial evidence from both fish and mammals has pointed to its presence and action in the ovary as a local peptide factor. The present study demonstrated that GnRH significantly suppressed the final oocyte maturation stimulated by all major maturation-promoting hormones or factors including DHP, hCG and activin. This suggests that GnRH may act directly on the oocyte to exert its action in the distal part of the pathway leading to oocyte maturation. Although the mechanism of GnRH action in the ovary and the regulation of its expression remain unknown, its potent inhibitory action on the oocyte maturation strongly suggests a critical role for it in the ovarian functions.
